バックグラウンド: | The Itg-Beta3 gene encodes the β subunit of the platelet membrane adhesive protein receptor complex Itg-Beta3 protein, which belongs to the Integrin class of cell adhesion molecule receptors that share a common heterotrimeric structure with a and β subunits. It is a common β subunit of the platelet complex and of two other Integrins (Fibronectin and Vitronectin Receptors), which have distinct a subunits. Itg-Beta3 has been implicated in a wide variety of functions, including platelet aggregation and thrombosis and implantation, placentation, angiogenesis, bone remodeling, and tumor progression. Integrin alpha-IIB/beta-3 is a receptor for fibronectin, fibrinogen, plasminogen, prothrombin, thrombospondin and vitronectin. Integrins alpha-IIB/beta-3 and alpha-V/beta-3 recognize the sequence R-G-D in a wide array of ligands. Integrin alpha-IIB/beta-3 recognizes the sequence H-H-L-G-G-G-A-K-Q-A-G-D-V in fibrinogen gamma chain. Following activation integrin alpha- IIB/beta-3 brings about platelet/platelet interaction through binding of soluble fibrinogen. This step leads to rapid platelet aggregation which physically plugs ruptured endothelial surface. |
