バックグラウンド: | The R-loop is a special chromosome structure that contains one strand of single-stranded DNA and another strand comprising a DNA:RNA hybrid. The distinct intermediate A/B conformation of the DNA:RNA hybrid makes it more stable than either dsDNA (double-stranded DNA, B form) or dsRNA (A form). R-loops normally form during transcription and play essential roles in key processes including chromatin modification, transcriptional regulation and termination, DNA replication and damage responses and genome stability. Most of the previous methods for genome-wide R-loop detection are based on immunoprecipitation using the monoclonal antibody S9.6 (which recognizes and binds to DNA:RNA hybrids in a non-sequence-specific manner) combined with high-throughput profiling such as tilling array hybridization and next-generation sequencing. |
